DIHYDROLIPOAMIDE ACETYLTRANSFERASE COMPONENT OF PYRUVATE DEHYDROGENASE COMPLEX (PTHR43178)
Sequences with problematic annotation (ID + gene/protein name):
Dmel Dbct (FlyBase=FBgn0030612|UniProtKB=Q9VXY3)
Human DBT (UniProtKB=P11182)
etc
Type of Issue: Erroneous source or erroneous propagation, or other issue
Both?
Fly Dbct (human ortholog = DBT) is the transferase component of the branched-chain alpha-ketoacid dehydrogenase complex and has the activity of GO:0043754/EC:2.3.1.168 (dihydrolipoyllysine-residue (2-methylpropanoyl)transferase activity).
Both these genes are the sole fly/human members of PTHR43178.
So there seems to be a a few problems here:
PTHR43178 is called DIHYDROLIPOAMIDE ACETYLTRANSFERASE COMPONENT OF PYRUVATE DEHYDROGENASE COMPLEX - i.e. it refers a different (albeit related) complex.
PTHR43178 is annotated with acetyltransferase activity, which is consistent with the Panther name, but not the function of the human/fly members. That annotation is based on two sources:
(ARATH|TAIR=locus=2083358|UniProtKB=Q9M7Z1)
this is Arabidopsis BCE2 (Lipoamide acyltransferase component of branched-chain alpha-keto acid dehydrogenase complex, mitochondrial)
UP annotates it to EC:2.3.1.168 (=GO:0043754), which is correct
But it has an EXP GO annotation to acetyltransferase activity, which is wrong - should be acyltransferase activity, or probably the much more specific GO:0043754
(ECOLI|EnsemblGenome=BAB96685|UniProtKB=P06959)
this is aceF (Dihydrolipoyllysine-residue acetyltransferase component of pyruvate dehydrogenase complex)
UP annotates it to EC:2.3.1.12 (= GO:0004742), which is correct
and it has an EXP GO annotation to GO:0004742 (dihydrolipoyllysine-residue acetyltransferase activity), also correct
Bottom lines:
PTN001899725/PTHR43178 needs splitting/revising to correct the name/members/GO-MF annotation
the TAIR annotation to BCE2 needs checking/revising @tberardini
