We have a collection of HTO libraries (TotalSeq-C) and would like to run a demuxing algorithm to call singlets/doublets on the barcodes.
Seurat's HTODemux seems to require the GEX data, and so does demuxEM.
Would it be possible to run the demuxEM algorithm without a GEX library? If we trust the list of barcodes from running CITE-Seq-count with cells=N, is there a way to run the demuxEM code without providing the RNA expression hdf5 counterpart to the HTO data?
This is for libraries where we did the 10X VDJ libraries, so we don't have a 5'GEX.
