A small test of estimating richness using R package
breakaway. I used Annelide OTU Olig01 for the test.Conclusion: practically the same result as we obtain via glmmTMB and using an offset term for number of reads. Therefore, no need to pursue this further. But anyway, now it is documented that this is tested. The rendered html is here.
